As regras são sempre as mesmas: os alunos têm de agarrar os ingredientes do distribuidor para completar as suas receitas, mantendo em mente as reações mágicas que você pode desencadear. Eles têm que fazer uso inteligente das poções e preparar durante o exame para fazer um trabalho mais rápido e melhor do que todos os outros. Potion Explosion é um jogo familiar, com uma boa parte de estratégia. Os jogadores podem escolher tiles de receita e comprar bolas coloridas de ingredientes a partir do distribuidor. Se eles fizerem bolas da mesma cor colidirem depois de tomar um ingrediente, esta reação mágica também lhes permitem agarrar esses ingredientes. Cada poção tem um poder especial que pode ser usado apenas uma vez durante o jogo; esses poderes lhe permitem criar combinações e provocar ainda maiores reações em cadeia, a fim de completar as suas receitas mais rápido. Cada poção vale pontos, com o valor sendo dependente de quantos ingredientes são necessários para completá-lo.
